If the messenger and receptor fit perfectly, no signal culd be sent. In the process of binding, the messenger isn"t changed 6. One side is the signal reciever, the other side is a kinase. Messenger needs to induce a conformational change that turns on the kinase. Some agonists are so potent we need a low amount; others are weakly potent. We have opiod receptors in brain that recept opiod compounds we make naturally. Opiods from the envuironment can mimic ours and thus bind to these receptors leading to the same effect.
