Allosteric Regulation
- Ligand binds to site other than the active site
- Binding can inhibit or activate the enzyme
- Phosphofructokinase
o Catalyzes (Fructose 6-phospate + ATP
Fructose 1,6-biphosphate + ADP)
o Important step in glycolysis: highly regulated
o It is inhibited allosterically by PEP
End product of the pathway
Feedback inhibition: as [PEP] increases,
the pathway shuts itself down
o Dimer of dimmers
o Active site residues contributed from both
dimers
o Binding of F6P hyperbolic in absence of PEP
however becomes sigmoidal in presence of
PEP
Phosphofructokinase:
Arg162 (+) needs to bind to F6P (-)
