• Formation of DSBs during meiotic recombination in yeast and human involves the type II topoisomerases Spo11 (iniatior of DSB in prophase I)  preferably in intergenic region o Can use ChIP-chip or ChIP-seq for comprehensive ID of hotspots o No recombination = non fertile gametes • Recombination between repetitive sequences may cause non-allelic HR and genomic reorganization – cancerous cell common • Site specific integration of the bacteriophage lambda relies on HR at equivalent att site in bacteria chromosomal DNAand lambda DNA • Site specific integration can create knockout strains of yeast • Cre recombinase system of PI bacteriophage has been widely adopted for recombineering in many model organisms o High affinity for lox site o The Cre-loxP system can be used to generate tissue specific knockouts IN VIVO!  CROSS CRE MOUSE WITH LOX SITE MOUSE • Transposons could mobilize either by conservative method or replicative method • Mobile genetic elements or transposons can be divided into two groups: DNA transposons (cut and paste) and ret
