Lab 6: mammalian in vitro cell culture, 2d cell culture vs 3d cell culture, the future is 3d cell culture. Adherent vs non-adherent cell culture: adherent, fibroblastic, adherent, epithelial like cells, non-adherent, lymphoblast like, don"t attach to a surface, cell culture can be liquid for cells that normally do not adhere to matrix surface. Cells that originate from a solid tissue adhere to a surface via adhesion proteins expressed on the plasma membrane. Adherent cell cultures: 2d vs 3d: 3d allows cells to attach to the matrix in all dimensions, more biologically relevant: no cell polarity. The dish can be coated with a collagen or bioactive molecule with ecm. Can also use natural materials to provide matrix: low cost, standardized protocols, reproducible. 3-d cell culture techniques: cells can grow in presence or absence of the scaffold material. In scaffold free techniques, adherent cells form clusters by attaching to each other via cell adhesion proteins.
