Cell biology test #1 stuy guide: bright field microscopy/ compound light microscope: protocol: 1. Choose a chemical that will cross-link tissues/cells (covalently bond them together), formaldehyde cross-links proteins. Cut sections using microtone, typically 10-15 micrometers thick slide. Replace with xylene, then replace that with ethanol, and replace that with water. With water, you can now stain the tissue. Alternative: to do it quickly (ex in an operating room) use cyrosections: freeze the sections and then just section it. Uses: to look at animal cells bacteria, mitochondria, and primary viruses. Typically used in pathology to help with diagnosing diseases: phase contrast microscopy: technique: degree of light/darkness depends on the refractive index of that region. Brighter if in phase, darker if out of phase. Used to observe single cells or thin layers. By stacking images taken at various points in the cell obtain a 3-d image a dic image is a thin slice through objects.
