Endocytic pathway: specific proteins of the membrane will form vesicle of internalization thanks to association with clathrin molecules. Goes from early endosome to late endosome to lysosome. App is transmembrane protein, synthesized in er then brought to plasma membrane. App retained at plasma membrane cleaved by alpha secratse, makes c83 fragment. App reinternalized through enodyctoc pathway, cleaved into c99 fragment in early endosome, then cleaved into ab in late endosome bad. App retained at plasma membrane: protective non amyloidegenic processing good. App internalized at full length: pathogenic amyloidgenic processing bad. Could affect trafficking pharmacologically so don"t have to mess with secretases. App localized to the plasma membrane, golgi, and endosomes. Detects full length app seen a lot at plasma membrane. App internalizes from the plasma membrane into intracellular compartments, endosomes, and golgi.
