Drugs, Lec 5:  01/27/2014 New compounds: NCE: new chemical entities: molecules never extracted from the environment or  created by synthetic chemistry. Don’t know what they’ll do to the physiology. No data on what it does in  vivo. Medical schools want to find out what it does.  Drug Dose­effect curve: Dose (x­axis) Effect (y­axis). Ex. interested in sedative­hypnotic NCE:  Effect % of rats  100  Asleep 30 mins  50    best fit sigmoid Post injection                       25      ED50=3                        0                    V        1       3   30    100                Dose (increases left to right) (mg/kg) Logarithmic scale on base 10. (If 1,2,3 additive, if 5,10,15 geometric). V doesn’t count.  Why logarithmic scale? We don’t know the potency! It matters.  V is vehicle. Vehicle is what you put the drug in to get it to the animal’s body.  You test all of the subjects on both doses. If one is 0% and the other is 100% take a half point (for logs it’s  about a third).  Biphasic drug effec
