Need to do other things other than regular nutrient broth. Know what you are trying to culture. Dilute first to rid of rare weed species. Select for oligotrophic microbes that grow slowly. And take out weeds that overgrow other microbes. Ex. second microbe that produces hydrogen which is needed by first microbe. Single cell in each well will be able to potentially grow w whatevver nutrients added. Have slow growing microbes => won"t be overgrown. Won"t be competing for nutrients in low abundance. Microfabrication techniques to construct thousands of indiv wells in small device. Only one cell can fit in well. Toward top of column = photosynthetic microbes. To look at morphology, help to see how many, Fluorescent staining using dapi, acridine orange (ao) or sybr green i. Cannot differentiate b/w live and dead cells. Viability stains: test to see if its live or dead. Take sample and see what"s actually there, no manipulation.
