28 September Substrate Level Phosphorylation Step 6 of glycolysis Net ΔG°’ = +.15 kcal/mol First payoff step Start with glyceraldehyde 3-phosphate Enzyme is glyceraldehyde 3-phosphate dehydrogenase Has sulfhydryl functional group Reacts through nucleophilic attack with carbonyl group Forms connection – covalent catalysis Covalent bond with C Use NAD to oxidize glyceraldehyde 3-phosphate Takes 2 e & proton Forms NADH (first product in payoff) High-energy bond between product and substrate Inorganic phosphate taken up Nucleophilic attack Displaces sulfur from enzyme – breaks enzyme away Results in 1,3-bisphosphoglycerate High energy Hydrolysis has –ΔG Step 7 Net ΔG°’ = -4.5 kcal/mol Starts with 1,3-bisphosphoglycerate Phosphoglycerate kinase transfers phosphate from 1,3-biphosphoglycerate to ADP → ATP and 3-phosphoglycerate ADP + Pi → ATP ΔG°’ = +7.3 kcal/mol 1,3 BisPG → 3 PG ΔG°’ = -11.8 kcal/mol net = ΔG°’ = -4.5 kcal/mol Coupling of the two reactions Steps 6 & 7 ΔG°’ = -3.0 kcal/mol The TCA Cycle Is the “hub” for metabolism in the cell The TCA cycl
