Continued: last time, we were looking at how the recombinant dna strategy can be used as a technique for synthesizing artificial human growth hormone (hgh). These libraries are made from all the mrna molecules present in a population of eukaryotic cells at a given time. Hgh is only expressed in cells of the pituitary gland. If you were to make the cdna library from (cid:373)us(cid:272)le (cid:272)ells, where hgh is (cid:374)ot e(cid:454)pressed, there"d be no mrna for hgh. When a gene is first transcribed, it results in an immature mrna. This is important because we are using bacteria to produce a human protein (hgh) from a human gene (gh1). Prokar(cid:455)otes (cid:272)a(cid:374)"t spli(cid:272)e out i(cid:374)tro(cid:374)s and would make a large, nonfunctional protein from the gene. We use another tool called a dna probe: a single-stranded dna fragment of known sequence that binds to complementary bases in a given sample of dna.
