Polarized Light Microscopy (PLM)
Materials fall into two categories:
o Isotropic demonstrate the similar optical properties in all directions
they have only one or singular refractive index
light passes through them at the same speed with no directional
restrictions
gases and liquids
o anisotropic optical properties that vary with the orientation of the incoming
light and optical structure of the materlal
about 90% of all solids are anisotropic
the refractive indices vary depending upon the incident light and the
optical structure
most synthetic fibers are anisotropic
polarized light microscope is used to study the interaction of plane polarized light with
anisotropic substances
light that vibrates only in one direction is polarized light
a polarized light microscope is like a compound microscope with the addition of a
polarizer, analyzer, and circular stage (allows the sample to be rotated)
polarizer a specialized filter that allows light through in one direction
o place under the sample
o vibration direction left-to-right direction (east to west)
analyzer a specialized filter aligned in the opposite direction to the polarizer
o located over the objective
o vibration direction (north to south)
