Enzymes are essential in the biotransformation process. Enzyme can change its state to work more efficiently. Occurs within days in response to need for greater enzyme efficiency. Occurs within a week in response to the need for enzyme efficiency beyond that achieved with activation. Once in constant state of enzyme production addiction. Change in enzyme state can affect breakdown of multiple drugs. Metabolic tolerance in different types of drugs. 1 carbamazepine (anti-seizure) and birth control (in addition to smoking or antidepressant) Metabolized by a common p450 liver enzyme. Metabolized by a common liver p450 enzyme. Pt: tolerance occurs from increases in enzyme efficiency metabolic tolerance. Drugs work really well for a little bit but then stop working, better to prescribe a different drug broken down by a different enzyme as opposed to a higher dosage. Factors that can alter the physiological effects of a drug include: The time elapsed since administration (latency, elimination life)
