To make sure the sir2 insert was picked up. Add only half of the buffer so it does not boil over. Smaller volume of buffer melts the agarose faster. Put the cap on loosely so that steam can evaporate from the bottle when you microwave (to melt the agarose) Don"t put it for 5 minutes straight, otherwise it will evaporate. Add the other half of buffer to rapidly cool the gel so you can pour it quicker. Add water to the side of gasket to help it slide easier on the platform. Add running buffer to the chamber and to cover the gel. Take the combs out after adding the buffer. The tip of the pipette should be above the well, and let the marker flow down into the well; the glycerol will push the marker down. You should see bubbles which means that the current is running.
