Document Summary
Main use: hypnotics, drugs that foster drowsiness and induce sleep. 4 categories: based on duration of action (dosage determines clinical use: ultra-short acting & short-acting very lipid-soluble, leave blood readily, pass. Bbbarrier in seconds: intermediate acting & long-acting readily absorbed orally, water-soluble, slower to leave circulation, bind with less fat, protein, muscle remain active, circulating, ex. History: adolf baeyer (1864) synthesis: made from urea and malonic acid. General ns depressants: loss of motor control, sleep, euphoria, sedation. Facilitate gaba"s inhibitory actions by increasing sensitivity of receptors: high doses mimic gaba inhibition activate cl- channels, hyperpolarization. Affinity for reticular formation block arousal. Reduced oxygen consumption by depressing medulla respiratory center. Limbic system (low dose) cause mild euphoria, disinhibition, increased aggressiveness (high dose) emotional withdrawal, depression. General uses: surgical anesthesia, anticonvulsant, detoxification, assisted suicide. Used as hypnotics used to be sleeplessness drug of choice, but although it allows initial sleeping, but decreases.
