November 22, 2013 Molecular cloning – recombinant DNA technology Plasmids (vectors) – small dsDNA that are physically separate from, and can replicate  independently of, chromosomal DNA within a cell Reproductive Cloning – duplicating the entire organism A plant or animal that has the same genes as the original from which it was produced  Sir John Bertrand Gurdon – first person to take DNA out of a differentiated cell and produce a  new organism  Somatic Cell Nuclear Transfer (SCNT) – embryos are made by combining the DNA of a somatic  cell with an egg cell from which the nucleus has been removed • Step 1: Enucleate (remove DNA) from unfertilized egg • Step 2: Combine somatic cell nucleus (DNA) with enucleated egg cell (creating a zygote) • Step 3: Allow zygote to develop until blastocyst stage (implantation stage) and implant  into mammalian surrogate uterus of the same species  DNA + protein complex = chromatin How is chromatin altered? • Chromatin­remodeling complexes (need ATP) • Histone modifications o HATs reduce the positive cha
