Lecture 8 Protein Folding 1. There are many different types of proteins (not just globular) 2. The reason that globular proteins fold is because of the enthalpic and entropic  preference Levels of protein structure 1. Primary (AA sequence) 2. Secondary (a­helix, b­sheet) a. 2.4 Super Structure (subset of secondary elements that go together) b. Hydrogen bonding between carbonyl carbon and the amino group on the  a­carbon c. S­S bridges i. This locks the polypeptide into a specific structure 3. Tertiary (Overall Fold in 3D) a. Location of all the 300 alpha carbons b. Formed by all the side chain interactions in the molecule i. Hydrophobic ii. Ion Pairing, Salt bridges iii. H­bonds (can occur with side­chain, OR main chain NH2, COO) iv. Water interaction c. 3.5 Domains  i. one part of the chain folds to create one structure ii. another part folds to make another structure (2 domains) iii. all the domains are connected together by a single amino acid  sequence iv. Titan is an example 4. Quaternary a. It is usually seen as many polypeptides bound together b. Hemoglobin c. Think hetero/homo dimers d. Ribosomes (complex of poly peptides) Fibrous protein and globular proteins do not function the same 1. One exists in the membrane 2. Another exists in the cytosol Hemoglobin & Myoglobin structure 1938 uncovered by x­ray crystallography 1. A lot of fibrous proteins were uncovered during this time Fibrous Proteins 1. a­keratin (nails, hair, horns, beaks, scales) a. made of alpha helixes b. an extended alpha helix (does not fold up)  c. large amounts of S­S bonds 2. B­keratin (silk, spider web, feathers) a. Beta plated sheets Lecture 8 b. Parallel or anti­parallel 3. Collagen (tendons, ligaments, sinews) a. Very stable proteins b. Most abundant 4. Actin & Myosin a. Muscle Fibers b. But these fibers are NOT made of fibrous protein c. Made of Globular proteins, assembled in a linear manner Alpha amalayse 1. G
