Document Summary

Ca2+/camkii - a calcium activated molecular switch for memory calmodulin can undergo. Key downstream component that turns on when there"s an in ux of ca2+ through nmda activators during induction of ltp. Activated by calcium/calmodulin => kinase activity shifts from inactive form where inhibitory domain is in contact with catalytic domain to active form. Camkii rst binds calmodulin before undergoing a conformational change => calmodulin is then removed, while kinase activity stays on. When ca2+ to returns to basal levels and camkii attempts to close, it can"t and remains on. Camkii cannot return to closed-state due to autophosphorylation. Transient increase in ca2+ can cause persistent activation of camkii. Robust kinase activity in absence of ca2+ Adding ca2+ will revert it back to fully activated state. Camkii is robustly activated following induction of ltp. After increase in ca2+, there"s a translocation of camkii to area where nmda receptors are located. Camkii binds to activated nmda receptors and binds to phosphorylated proteins.
