PHRX 4040 Lecture Notes - Lecture 3: Ventrolateral Preoptic Nucleus, Dual Role, Prostaglandin
Document Summary
Ach - not too much of a role, drives wakefulness. 5ht - (more in next slides) ie) amphetamines increase catecholamines. H - very important in wakefulness (more later) ie) sleepiness from antihistamines (h1 that cross bbb) Orx - extremely important. most active during waking/motor activity. Il and immune modulators, prostaglandins (d2, e2), endogenous sleep factors a) b) In animals, not well characterized, lack drugs ie) body temperature and prostaglandins. Secretes gaba to inhibit activity of wakefulness areas (aras) Most sleeping drugs work through gaba (don"t mind nuclei and details on slide 24: homeostatic sleep factors - need for sleep. Unopposed = sleeps in short bouts through day. Maybe different receptors, may depend on time of day. You"re awake now but will have to sleep eventually . Primary = promote wakefulness, 5 ht stim. Secondary = also causes synthesis and release of sleep-promoting factors. Sleep switch activated once threshold is passed (vlpn) Studies: increases sleep in animal studies, caffiene = antagonist.
