Medicinal chemistry is interdisciplinary science that touches upon: chemistry, life sciences, physical sciences. What is a drug: compounds which interact with a biological system to produce a biological response. Cannot kill all known bacteria, resistant bacteria, many have allergic reactions: morphine. Result in tolerance, addiction, respiratory depression, deadly overdose. Drug of choice for treating patients dying of cancer to reduce pain. Therapeutic index: measure of drug"s beneficial effects at low dose versus harmful effects at high dose, td50/ed50. Selective toxicity: toxic against foreign or abnormal cells but not against normal host cells. Antibacterial, antifungal, poisons to microbial cells but not human cells. Anticancer poisons to cancer cells not human. Antiviral agents: important for drugs to be clinically effective. Drug definition by fda: effective for approved condition, safe, stable, deliverable and bioavailable, available, novel, patentable. Drug targets: specificity of drugs is a result of where they act in body- drug target, most targets are on cell membrane.
