Anxiolytic effect - inhibition of neurons in stress circuit (i. e. bla) Almost all neurons in the brain have. Act on gaba a receptors make more efficient in opening the channel (more inhibition) Prolong the synaptic actions of 5-ht at their receptors by inhibiting reuptake. Anxiolytic actions not immediate develop slowly over a period of weeks. May initially increase anxiety (promotes activation of certain crf neurons) Immediate rise in extracellular 5-ht not responsible for the anxiolytic effect. Effect = adaptation of brain to chronic elevated [5-ht] in brain. Ex: increase in glucocorticoid receptors in the hippocampus. Chronic activation of 5-ht 2 receptors causes up-regulation of glucocorticoid receptors in the hippocampus.
