Document Summary

The plasmid that we transformed consisted of: Bla = destroys the essential structure of penicillin family of antibiotics. Arabinose was regulating the gene expression on our bacteria. Gene expression is not efficient, and therefore heavily regulated. Non-fastidious = not picky bacteria that can grow in nearly any basic culture. If there"s a mix of sugars in the culture, the growth graph looks like two s-curves connected. The bacteria went from using one sugar to using the other. To determine what sugar is being consumed first, vary the concentration of sugar to something other that 50/50, and look at the growth curve. If there was no regulation, then there would be one smooth s-curve. Replica plating is like creating a stamp and an identical plate of bacteria that you want to study. Good for looking at cells with certain mutations. Cells that cannot take up lactose = permease-minus.
