Moa: binds competitively to ergosterol (in fungal cm, not humans), Perm/ leakage of k = cell death (also toxic to us b/c it binds to sterols in ours, but it binds better to ergosterol): amphotericin b: amphoterrible! Moa: azoles inhibit the conversion of lanosterol to ergosterol (which is needed in the cm). Permeability/leakage of k, inhibited cell growth/replication: imidazoles: topical infections, tinea. Drug interactions: drugs that acidity ( h2 antagonists, antacids, cyp450 interactions ( concentration of other drugs, triazole: newer ones, cyp 450 interactions. Fluconazole: diflucan, is safe and has a long half life; hence the 1 dose. Side effects: ** well tolerated: gi, liver functions: increases tests, liver necrosis = rare.
