Set up the cross into four equal quadrants (tan and wild opposite one another) Scalpel used to cut small cube of agar carrying fungal hyphae from either tan and wild plates collected and placed on corresponding quadrant of the plate (2 x tan + 2 x wild) Cubes placed close to centre of the quadrants, plate is sealed to prevent dehydration. Incubated at 22 degrees celsius for 14 days (allowing the fungus to grow). Organisms composed of cells (further composed of organelles, membranes and other structures) Whole cell movement: locomotion of the entire cell through flagella/ cilia or amoeboid movement. Intracellular movement: distribution of intracellular contents through cytoplasmic streaming. Demonstrates the presence of organelles and other intracellular structures that move. Organelles: identification of major sub-cellular organelles visible using light microscopy. Imaging techniques: light microscopes have a shallow depth of field, and the selected focal plane influences the image produced. Organisms studied: sperm from ram: haploid male gamete.
