Lecture 5 The Value of ATP - Hydrolysis of ATP ADP + P provides free energy that can be used to drive coupled reactions What is ATP? = Adenosine triphosphate - has two phosphoanhydride bonds formed between two phosphoryl groups accompanied by the loss of water - ATP has features that cause it to have high free energy compared with the hydrolysis products ADP & AMP: o Charge Repulsion the triphosphate is highly negatively charged under physiological conditions (pH 7.4) so charge repulsion is high. There is LESS charge concentration when these phosphates are separated. o Hydration  the ATP hydrolysis products are more easily solvated in water than ATP. Negative charges are separated and there is greater disorder o Resonance  in the phosphoanhydride bonds there is competition between neightbouring phosphoryl groups for the O electrons, resulting in resonance (alternative structures that differ in the position of electrons) - Considering these structural features together results in substantial phosphoryl group transfer potential ATP currency in the Cell - ATP drives several types of reaction
