Bred a mouse that would have allele a and allele b: they"re codominant, so it expresses both. They then took a true a and true b and irradiated them, which kills everything dividing quickly, including the stem cells in the bone marrow > kills leukocytes. This gives the cells that you put in a house to live in and makes sure that there are no previous immune cells. Every other cell that"s still alive in the mouse is specific for whatever it was. Now we put in bone marrow from the axb mouse into each the a and b mouse and measured the t cell response to presentation with apcs in vitro. It obviously happens outside the bone marrow, because the bone marrow has been transplanted. They found that cells from the animals that were exposed to the antigen that they had would respond to that one, despite the fact that they had bone marrow for both.
