The proper trafficking of lysosomal hydrolylase (lh) enzymes to the lysosome requires the presence of one or more mannose 6-phosphate (m6p) residues in their n-linked oligosaccharide chain(s). The phosphorylation of the mannose sugars on lysosomal-destined proteins is mediated in part by the cis-golgi enzyme n-acetylglucosamine phosphotransferase (glcnac-pt). M6p then serves as a signal patch that is recognized by the m6p receptor, a membrane protein found in the trans-golgi network, which also mediates the subsequent packaging of the m6p-bearing lysosomal proteins into clathrin-coated transport vesicles. You construct four plasmids expressing gfp-tagged wild-type or three different mutant version of an lh enzyme in order to study (via confocal laser-scanning fluorescence microscopy) its intracellular trafficking.
