Disruptions in a handful of key pathways are common to many cancers cancer-critical cells can go in 3 key pathways. 1% of genes in the human genome are cancer-critical. Mutations in the pi3k/akt/mtor pathway drive cancer cells to grow. The pathway starts at the membrane with a receptor tyrosine kinase. This leads to activation of a lipid kinase called pi3 kinase. Pi3 kinase causes production of pip3 and this leads to production of kinase called akt. Akt further activates mtor and mtor is central to pathway bc it increases the rate of protein synthesis. You need new protein synthesis in order for cell to increase mass. mtor is also very important for the increase in glucose uptake which is apparent in cancer cells. Cancer cells take up glucose 100x more than normal differentiated non-proliferating cell bc mtor increases expression of glucose transporters. This pathway is important for also for synthesis for other molecules like lipids.
