BIO314 Laboratory in Cell and Molecular Biology Exam Notes • Types of Centrifugation- o Differential- separation based on the size of the particles and the number of g’s applied o Differential pellet- larger particles sediment faster o Sedimentation rate can be increased by the increasing the centrifugal force o Particles could undergo contamination and poor recoveries due to the presence of other particles- for instance, smaller slower travelling particles could interfere with the larger faster travelling particles o Rate- Zonal- separation based on the mass and size of the particle o Particles sediment in the zones containing other particles of similar size o No contamination because the sample is layered in a narrow lane on the top of the density gradient= so, faster and slower particles do not interfere with each other’s sedimentation o Isopycnic- separation based on buoyant density of particles o Density of the gradient must be greater than that of the particles- no pellet forms at the bottom, instead, particles travel until their density is the same as the density of the gradient.
