Let"s say, we have mixture solutions that contains three compounds (a, b and c) and we want to determine the concentration of compound a. We can do this by various methods such as spectroscopy but let"s assume that compound a does not have a signal unique then the other two compounds. In that case, we will have to separate compound a from the mixture and then calculate its concentration or whatever we need to do. This can be done by chromatography which is a separation technique. In this unit, we will learn bout the different types of chromatography and how each one can be used to determine the components of the analyte from the mixture. Chromatography is a physical means of separation and the analyte of interest is separated by phases. The physical basis of separation is the partition coefficient (k).
