RNA polymerase is tethered until TFIIH acts ---> it is a protein
kinase which attaches phosphate groups on to the carboxyl terminal
domain of polymerase II
o
After addition of the phosphates the other transcriptional
machinery will escape, and this causes conformational changes in RNA
polymerase II thus making it easier to transcribe
o
mRNA is made of 5' cap and a poly A tail
The 5' cap means that 7th position of guanine is methylated
and the 2nd position of ribose is methylated as well
Intron Splicing:
The introns are spliced out of the DNA, and the primary transcript is formed
Processing takes place such that a poly A tail and 5' cap is added
Genomic clone-
contains both the introns and exons
Complementary DNA clone (cDNA)- made from mRNA thus lacks introns
