Transfer the ovaries to the third well in your three-well glass dish, leaving the rest of the fly behind in the second well. Try to collect the ovaries from at least 3-4 females. Make sure you tease the ovarian tissue apart since there is a transparent ovarian sheath covering the ovaries that will impede. X-gal uptake and therefore staining: pick out females, as you are going to isolate the ovaries.
