B Lymphocyte Signalling – Igβ and Igα are req’d for surface expression of immunoglobulins B cell activation – B lymphocyte antigen receptor complex (BCR) = surface Ig + Igβ + Igα  binds antigen – B cell receptor clustering = signal passed into cell  Up-regulate antibody production  Starts secretion  Isotype switching stimulated by IL-4 or IFNs – BCR bound to antigen is internalized  endosome  Processed for MHC class II presentation  MHC class II w/ antigen put on surface of B cell  activates T cells  T cells secrete more cytokines  activate more B cells Inhibitory signals  ITIM: immune-receptor tyrosine-based inhibitory motif – [I/V] X Y X X L  X = any aa, Y = tyrosine, I = isoleucine, V = valine, L = leucine – SH2 domains bind phosphorylated tyrosines   ITIMs recruit SH2 domains contain phosphatases
