Passage through the cell cycle require serine/threonine protein kinases that are dependent upon regulatory unit known as cyclin: different partners of cdk/cyclin complexes mediate passage through these different phases of the cell cycle. We talked extensively about mpf (mitosis promoting factor) its kinase activity is required for every single eukaryote to go from g2 to m-phase. This is regulated via phosphorylation (inhibitory phosphate on y15) by wee1; and then the inhibitory phosphate is taken off by cdc25 phosphate (this is what triggers the entry into m-phase) If you have antibody against cdk2, this will inhibit cdk2 and prevent the cell from going into s-phase. Restriction point: the point which you no long need growth factors in the cell to proceed through the cell; if you take away the growth factors before this point and the cell will arrest there. Cyclin d expression is required for the passage through the restriction point.
