Lecture #5: Aconitase
What would happen to the activity of aconitase if we changed the pH? Why?
Specifically refer to mechanism and amino acid protonation status. Consider pH 4, pH 7, pH 10
As you raise the pH, deprotonation occurs and it becomes more negative (positive charge
slowly goes away). Interferes with the way it reacts with other molecules
Hydrophilic interactions increase the polarity
pH 4: histidine will be acidic and there will be a lot of protons around; will still have a
positive charge around it and act as an acid. With all these protons, the serine will be
deprotonated.
pH 7: most favorable condition for this activity; will be active; will probably be
protonated in order for it to react
pH 10: There will not be 2 protons around to protonate acids; will not be protonated; will
not go back to an acid; will not be active
