BOAG CHAPTER 18 Polymerase chain reaction (PCR).During each PCR cycle, short DNA sequences that are complementary to the ends of the targeted DNA sequence bind to the DNA and act as primers for the synthesis of this DNA region (see small illustrations next to arrows in cycle 1). The primers used in actual PCR experiments are usually 20 to 25 nucleotides long. The region between the two primers is typically hundreds of nucleotides long, not just several nucleotides as shown here.
