Document Summary

Cmv promoter is used because it helps make the dna (lysostaphin dna) We want a poly a signal in the expression in our expression so that it enhances translation, increases rna stability, and its last role. The components you need are promoter, the dna of interest and poly a tail. The kozak site- is needed if you want to initiate translation of the bacterial protein. A membrane protein already has a similar site. The vector (3) is our main construct they also made another vector (2) which has the lysostaphin replaced by the hgh (human growth hormone) which is totally unrelated protein compared to lysostaphin. They preform western blot, from samples from different places. One is the cell extracts and the other is the medium. You can see that they use ab against the lysostaphin, in lane 1 (is a possible control) they cultured the lysostaphin producing bacterial.
