Neurotransmitter system includes: neurotransmitters, molecular machinery to make the neurotransmitter, package it into vesicles, participate in reuptake and degradation as well as the chemicals involved in the transmitter"s action. Cholinergic: the neurons that make and use ach. Ach used in a cholinergic system, ne in a noradrenergic system. Must prove that it"s localized and made by particular neurons. Immunocytochemistry: use to localize molecules to particular cells. Purify the candidate for a neurotransmitter and inject into blood of animal cause immune response. Antibody bind to transmitter candidate, recover and tag the antibodies. Put tagged antibody in brain tissue and it should bind to transmitter in question only. In situ hybridization: confirms that a cell makes a certain protein/peptide. Create a complementary strand to the mrna making the peptide in question (called a probe) Go through hybridization where the probe binds to the mrna strand and then chemically label the probe.
