Biochemistry 2280A Chapter Notes - Chapter 12: Nylon, Intron, Stem-Loop
Document Summary
Sequence of dna must be constructed from series of shorter sequences. Shotgun method: dna broken in to small fragments, computer used to search for regions of overlap. For a eukaryotic genome there are 2 methods. Gene map: provides a guide for the sequencing experiments. Shows positions of genes and other distinct features. Whole -genome shotgun method: same as shotgun sequencing but uses gene map as landmarks to help build master sequence. Clone contig method: genome broken into small segments, short enough to be sequenced by shotgun method. Then positioned at correct location on genome map. Genetic mapping: use genetic techniques to construct maps showing positions of genes and other sequence features on genome. Physical mapping: examine dna directly to construct maps showing positions of sequence features. Clone contig method: genome broken into segments with known position on genome map, before sequencing is carried out.
