Histology is the bridge between structure and function! Stains are used to provide contrast, so we can see the specimen! Typically, hematoxylin stains nucleus and eosin stains cytoplasm. Less speci c: cytoplasmic lament, membranous components, most extracellular bers! The structures that have a high affinity for the h dye are most heavily stained. e. g. nuclear dna and areas of the cell containing cytoplasmic rna. Periodic acid-schiff (pas) reaction stains carbohydrates and carbohydrate-rich macromolecules. It is used to demonstrate: glycogen in cell, mucus in certain cells, the basement membrane that underlies epithelia, reticular fibers in connective tissue kidney tissue stained by the pas method.
