Epigenome • Telomerase is normally turned off in somatic cells • Reactivation of telomerase can lead to aging cells that continue to proliferate, a feature of many types of cancer • TERT reactivation is one of the most common mutations in cancers of all types • Histone proteins in the nucleosomes can be modified by adding or removing chemical groups • This affects the strength of nucleosome-DNA associations, and can cause chromatin structure to form open promoters, or condense, leading to closed promoters • Principal chemical modifications are addition and removal of acetyl and methyl
