MCB 2610 Chapter Notes - Chapter 1B: Electron Microscope, Flagellum, Electromagnet
Document Summary
Avery welch mcb 2610 microbiology week 2 & 3 chapter 1 appendix b microscopy & specimen prep: simple stains. A single stain is used (easy and quick) Can determine size, shape, and arrangement of bacteria. Differential staining: divides microorganisms into groups based on their staining properties. Divides bacteria (but not archaea) into 2 groups based on differences in cell wall structure. Steps: make a smear of a bacterial suspension on a glass slide. Avery welch mcb 2610 microbiology week 2 & 3 chapter 1 appendix b microscopy & specimen prep. Particularly useful for staining members of the genus mycobacterium (which don"t stain well with the gram stain) High lipid content in cell walls (mycolic acid) prevents dyes from readily binding. Uses high heat and phenol to drive basic fuchsin into the cells. Acid-fast positive is red & acid-fast negative is blue. Opposite of gram stain: differential staining of specific structures.
