Document Summary

Chapter 6 genetic analysis and mapping in bacteria and bacteriophages. Genes are arranged in operons a group of genes that are transcribed together because they have interacting functions. Selective media used to find mutants. Bernard davis (1950) discovered that physical contact was required for genetic transfer. Fertility factor (f factor): donor cell, conjugation pilus, recipient cell. Bacterial genetic material transfer done by: physically linking together and transferring dna, requires f plasmid, one-way transfer. F plasmid: uses dna replication machinery of the cell, inhibits contact with other f+ cells, has genes that promote the growth of pili. Note: when the pilus contacts the donor cell, the strand to be transferred (t strand) is cut by cleaving a phosphodiester bond. Homologous recombination - can occur in a merozygote and therefore transfer genes between bacterial chromosomes: transfer of donor fragment sets up the opportunity for recombination of chromosomes, recombination between donor and recipient occurs.
