A protein assay is a way of detecting for the presence of a specific protein and estimating the concentration of the protein. The assay should be specific to the protein you are studying and based upon unique characteristics. Enzymatic activity, immunological assay (antibody) and biological activity/function. Select a source of your protein carefully based on the source of the cell, opting to find one easily obtainable in large amounts. If you are studying a muscle protein, you would use muscle tissue as the source. Other factors to consider are low concentrations of proteins that may co-purify, and low concentrations of proteases that may destroy the protein of interest (you can add protease inhibitors to combat this problem) The best solution may be to express your protein in an alternate cell type. Step 3 - break open cell (protein extract) If you are studying a cytosolic protein or a secreted protein, you would expect that it would be soluble.
