Document Summary

Toxicology: adverse effects of chemicals, physical agents on living organisms, assess probability of hazards, estimate result of effect on populations, define qualitative, quantitative exposure response relationships. Identify factors that increase/decrease susceptibility to toxicity in individuals/populations: develop prevention/treatment methods. Toxin: produced by biological system (plants, animals, fungi, bacteria) Mechanisms of toxicity: via reversible binding of parent molecule/stable metabolite to cell receptor. Excess binding to intended receptor od. Bind to unintended target: bioactivation of relatively non toxic chemical reactive metabolite covalent binding/oxidizes dna/protein/lipids. Duration, frequency of exposure in animal studies: acute <24 hors, ip, iv, sc, oral intubation, dermal (different routes, single administration, subacute repeated, <1 month, subchronic repeated, 1-3 months, chronic repeated, 3+ months. Exposure assessment: lab observations vs. real life (extrapolation is a challenge, direct vs. indirect (potential biomagnification) chemical measurements. Dose-response assessment: can be many dose-response relationships for each chemical, graded vs. quantal dose-response, response of individual, response of population.
