Document Summary

Serum iron, total iron binding capacity, serum ferritin. Total iron binding capacity a measure of transferrin concentration. Ferrozine absorbs strongly when it binds iron. Ferrozine will only bind iron not bound by transferrin. Ferrozine will not bind ferric iron- ferric iron is the form of iron bound to transferrin. Use a buffer with a ph of 4. 5 containing ascorbic acid (vitamin c) Low ph causes iron to dissociate from transferrin. Ascorbic acid reduces ferritin iron to ferrous iron. Quantitate using a standard of iron in solution, using the formula in the lab manual. Total iron binding capacity (tibc) is a measure of transferrin. Tibc increases w/ iron deficiency, as the liver synthesizes and secretes more transferring into the plasma. Tibc will also be measured using a dye that binds iron -chromazurol b. First step-- add low ph solution of iron-binding dye and iron to serum. Low ph buffer contains ferric chloride and iron-binding dye.
