Document Summary

At end of gene, poly-a signal sequence in dna is transcribed into rna, triggers transfer of cstf and cpsf from rna pol to rna. Enzymes (factors) switch from being associated with polymerase to rna. Rna is cleaved off rna pol ii, poly-a tail added and termination of transcription. Poly-a binding protein binds to poly a tail to stabilise the rna. Poly-a tail is further extended in the last phase. Length of poly-a tail is not random and important for mrna stability and function. 3 rna polymerase enzymes (only 1 in prokaryotes) Primary transcript = prerna = heterogeneous nuclear rna (hnrna) Guanyl transferase adds a guanine nucleotide (7mg/7-methylguanosine)to the 5" end of the nascent pre-mrna (while still being transcribed) 5"-capping is initiated once 20-30 nucleotides have been transcribed. 3" modification: polyadenylation poly-a sequence added by a poly-a polymerase. Pre-rna (still with introns) contains consensus nucleotide sequences that signal the beginning and end of introns.
