Identify all regions of transcription, transcription factor association, chromatin structure and histone modification in the human genome sequence. Output = a complete functional map in different tissues and at different times - functional characterisation of the human genome. Also have a small amount of dideoxyribonucleosides (a, t, c, g) We extend primer and we get a range of fragments of different lengths. Acrylamide gels used as they can resolve one bp difference. Automation of sanger sequencing was a key enabling technology for the human genome project. Scan with a laser that detects the light and allows us to determine sequence. Ngs: fragment genome into much smaller fragments (500bp). We sequence the fragments at the same time.
