BIOM30001 Lecture Notes - Lecture 15: 3D Cell Culture, Xenotransplantation, Collagen
Document Summary
Series of mechanical, enzymatic and/or chemical treatments to remove cells from an organ. Provides an acellular, naturally occurring 3d scaffold of ecm. Ultra-structure and arrangement of native and complex ecm are conserved. Collagen rich matrix to support growth of selected seeded populations. Dynamic interactive environment between cells and tissue specific ecm. Topology and ecm profile most closely support recapitulation of the organ, immediate vascularisation and integration with surrounding tissues. Ecm is highly conserved across species and will therefore be well tolerated across individuals. Could use an animal ecm populated with human cells. Atm tissues produced are functional but only survive for a few hours. Allogeneic-use organ donations only (cid:858)perfect(cid:859) organs used for transplant remainder could be utilised for scaffolds. Techniques for the manufacture of scaffolds are becoming more sophisticated including computed assisted 3d fabrication of custom scaffolds and tissues ie. robotic printing. Delivery of controlled gradients of biologically active factors. Can make complex tissue with high precision in hydrogel moulds.
