Patients vary with respect to many factors that can affect the response to a drug. For ex: the patient"s sex, age, body composition, rate of drug metabolism, and the rate of renal excretion can affect both the pharmacokinetics and pharmacodynamics of drug response. Disease states and co administrations of other drugs can also affect how a patient responds to a drug. During phase 2 trials, volunteer patients are given a range of doses beginning with a very lose dose to determine the immediate and delayed pharmacologic response. Peak of the frequency distribution curve represents the dose that produces the desired therapeutic response in 50% of the sample. Peak is called the average effective dose, or the ed50. Dose that would be selected for initial treatment in phase 3 trials. Phase 3 trials dose would be titrated up or down according to the patient"s response.
