Endoplasmic reticulum is an extension of the nuclear envelope which surrounds the cell. Golgi contains stacks called cisternae, cis- (near er) and trans- (away from er) Lysosomes contain hydrolytic enzymes that dispose of waste (sometimes called vacuoles) Distribute proteins, lipids and, metabolites throughout the cell - secretory pathway. Gfp - can be expressed in so many cell types, originated in jellyfish. Temperature-sensitive vesicular stomatitis virus g protein (vsvg) secretion. Temperature sensitive - at 40 degrees, it misfolds and accumulates around er. So this so it accumulates in the er, doesn"t move out - need this to happen so it can be visualised. At 32 degrees, it fold properly - allows you to visualise bulk movement of protein through endomembrane system. First accumulates (when its soluble) moves straight to golgi. Little vesicles bulge from golgi - then they migrate from golgi to the surface of the cell where they usually go. After 3 hours they are spread over plasma membrane.
